After upgrading to 4.3.0.30 a few of the Library items appear to have reappeared on the home screen.

I wanted to get down to 2 rows, so tried to switch off the Camera Uploads as I don't use it, but even with the checkbox unchecked I'm still seeing Camera Uploads on the home screen.

Checked and this is the same on aTV and iOS clients, as well as the HTML page.

Try refreshing the page or restarting the app.

That only applies to latest media and continue watching.

So there is no way of removing the Camera Upload button from the Home Screen?

Regarding the settings, for the next release I've updated the option text to:

Display in secondary home screen sections such as latest media and continue watching

You can use any of the existing user access features to restrict access to that library.
