I was thinking of using direct play, but I was wondering, everything on Synology is based around permissions. What permissions are required for an Emby client to directly play a file?

? The same permission required to transcode it.  I'm not sure direct play means what you think it does.

Doesn't it mean, for example, Emby client running on Roku can play/access the video file without going through the Emby server?

Correct. Roku will stream through Emby server.

So in order for Roku to have direct access to that folder/files doesn't it need some kind of login details or permissions?

There is nothing you need to do in order to be able to direct play on Roku. Just press play and enjoy your media. Please let us know if this helps. Thanks.
